Purified hematopoietic progenitors were transduced in the presence of IL-3, IL-6, and SCF, then plated into methylcellulose cultures, and GFP hematopoietic colonies of various types or morphologies colony-forming units mix [CFU-mix], colony-forming units granulocyte-macrophage [CFU-GM], etc. ; were picked at days 5 to 6 and plated into secondary methylcellulose cultures. Individual secondary colonies were then scored for GFP expression by FACS and for the presence of vector genome in the daughter cells by PCR. Within the 30 secondary colonies that derived from 7 different primary GFP colonies, only 42% were GFP positive by both FACS and PCR Table 1 ; . None of the colonies were positive by FACS without being PCR positive, demonstrating the high sensitivity of the PCR assay. Approximately half of the colonies 46% ; were negative both by FACS and by PCR. Some of the colonies 13% ; were positive by PCR without demonstrating any expression of GFP by FACS, representing either integrated copies where the expression from the EF-1 promoter was silenced or cases where the GFP gene was amplified by PCR from a nonintegrated vector. The presence of GFP PCR daughter colonies in the progeny of primary GFP colonies indicates that the lentivirus gene transfer is not fully completed between the time of transduction and the time when the transduced progenitor divides. Therefore, the vector genome seems to integrate after proliferation in the methylcellulose culture starts, and as a consequence, vector integration is seen in only a portion of each progenitor's progeny cells. Activation of the complement response following baculoviral infection has been investigated in more detail and it appears that complement is being activated via the classical pathway since serum depletion of C1q, unique to the classical pathway, allowed complete survival of the baculoviral vectors in vitro Hofmann and Strauss [78] ; . Hofmann and Strauss Hofmann and Strauss [78] ; explored various strategies for complement inhibition to promote baculoviral vector survival in vivo. Incubation of human serum with an antibody against complement component C5, involved in both classical and alternative pathways, promoted vector survival in a dose dependent manner in vitro Hofmann and Strauss [78] ; . In addition, treatment of human blood and plasma with cobra venom factor CVF ; , an inhibitor of the complement component C3, also resulted in almost complete survival of baculovirus vectors as opposed to the 1% that survived in the absence of this factor Hofmann and Strauss [78] ; . CVF has successfully been used to deplete the complement response in mammals in vivo, including monkeys Chen et al [79] ; . A recombinant soluble complement receptor type 1 sCR1 ; lacking transmembrane and cytoplasmic domains has been shown to inhibit both classical and alternative pathways of complement activation. Presence of this factor promoted baculoviral vector survival following incubation with human serum and subsequent transduction of Huh7 cells was approximately 5 times more efficient than with baculoviruses incubated with serum in the absence of sCR1 Hofmann et al [80] ; . A further strategy designed to inhibit complement activation by baculoviruses has been to engineer expression of decay accelerating factor DAF ; , a naturally occurring negative regulator of both classical and alternative complement pathways, on the virus surface as a fusion protein with gp64 H ser et al [71] ; . Following incubation u with human serum, the DAF modified baculoviruses were shown to be able to transduce Huh7 cells in the presence of complement at a much higher frequency than unmodified viruses. This was shown to be due to increased survival of the vectors in the presence of complement H ser et al [71] ; . The same vectors were injected into u rat livers in vivo and transgene expression, measured after 3 days, was five times higher in the livers treated with DAF-modified baculoviruses than the unmodified form H ser et al [71] ; . u Thus for cancer gene therapy, the complement response against Bv can be overcome by short term biochemical manipulation during viral inoculation. The optimal conditions have not been determined however, and a number of alternative inhibition systems, to those employed above can still be developed, particularly for prostate cancer patients. In the brain however, the complement response has no effect on Bv gene transduction, and it has been proposed as a safe and effective.

Possible to demonstrate that SDZ IMM 125 induced apoptosis according to the classical criteria, and to distinguish the effects observed from necrosis. Apoptosis appeared as typical cell shrinkage accompanied by condensed cytoplasm with focally dilated endoplasmic reticulum, structurally intact mitochondria and other organelles compacted together and maintaining their integrity, formation of cytoplasmic protuberances on the cell surface blebbing ; and cytoplasmic bodies, nuclear shrinkage with invagination of nuclear membrane, and massive deposits of chromatin in compact masses adjoining the nuclear envelope. It was also possible to clearly distinguish necrosis, characterized by swelling of all cytoplasmic compartments, swelling and disappearance of mitochondrial cristae, disintegration of membranes and accumulation of large dense granules in the mitochondrial matrix, detachment of ribosomes from rough endoplasmic reticulum membranes, and nuclear swelling with clumping of loosely textured nuclear chromatin Wyllie et al., 1980 ; . LDH release might be a component of apoptosis in cell culture. Although LDH release in cell culture does not necessarily imply necrosis, plasma membrane damage is one feature of necrotic cells, which could serve among other parameters to determine necrosis. Because under in vitro cell culture conditions the apoptotic cells can not undergo rapid phagocytosis as in vivo in the intact tissue, it might also be that LDH release is a result of late apoptotic cells. The fact that SDZ IMM 125 can induce necrosis was clearly shown by TEM examinations using morphological criteria 20 h after incubation. There were also clear indications for transition figures, showing cells with both apoptotic and necrotic properties. Instead the specific content and "meaning" ; of tourism have depended on its position within a larger framework of social interdependencies. Figurational sociology thus questions the ontological foundation for delimiting leisure as a specific form of practice. Instead of viewing leisure and tourism practices as compensation figurational sociology thus argues that leisure is essential mimetic Elias & Dunning 1986 p 66ff ; , and as such also an activity subdue to the transformation and variety of the social figurations their practitioners make part of, be that through work or private life op. cit & Rojek 1992 ; . Leisure therefore may exist in a "peculiar ambiguity", however this ambiguity is not necessarily a matter of alienated everydaylife versus the need for authenticity, but rather an ambiguity between the experimental opening of new horizons of experience and releasement of social restraints at the one side and social acceptance at the other. In this way the specific forms of practices related to leisure is a matter of negotiation Elias & Dunning 1986; p 90 ; . However figurational sociology by and large have neglected how specific forms of leisure practices are being negotiated. The focus have instead been on the historical production of forms of leisure in particular sports ; and their attachment to the civilizing proces .3 The emphasize put on the negotiation of leisure practices are also at the heart of the interactionist approach to leisure and tourism studies. This third way of contesting the cultural studies approach takes it starting point in an objection towards the tendency to equalize tourist experiences and tourist intentions - thus leaving out of focus the essentially negotiated character of tourist experiences. Tourism experiences are produced through complex social encounters between a variety of agents of which "the tourist" is only one, and subsequently essentially a negotiated experience. This is the central argument of Crang discussed earlier. Whereas Crang points to the rich sources of inspiration made up by interactionist social theory Goffman, Giddens and de Certeau ; . Empirical work on the negotiation of tourist experiences have been very sparse. One outstanding exception here is Edensors 1998 ; studies of negotiation of tourist experiences and practices. As is clear from Edensors ethnographic study not only the place myth and expectations attached to the same destination differ between various social and ethnic groups, so do the "walking practices" deployed by tourists ibid. p 105ff. see figure p 108 ; . I will return to this latter position, however I would like to emphasize that in spite of the differences between the four different apprioaches discussed here, I do not they are exclusive. To the contrary the three corrections of the cultural studies approach to the study of the sociology and geography of tourism. In the next section, I would like to return to the question of space and place in tourism.
