The results of this study show that PIXY321-mobilized CD34 + cells produced more MKs than GM-CSF-mobilized CD34 + cells, while GM-CSF mobilized more CD34 + cells per MNC collected than PIXY321. Together, this led to no net difference in the number of MKs produced per MNC cultured. One explanation for this finding may be that PIXY321 mobilizes a different subpopulation of CD34 + cells than GM-CSF, with a greater commitment to the megakaryocytic lineage, but is a less effective mobilizer of hematopoietic progenitors overall. In view of the small number of positive cells in certain samples, we ascertained the statistical validity of our results in three different ways. First, the CD34 frequencies were confirmed by another laboratory that analyzed the same patient samples for another study. While their sample preparation protocol was different from ours in particular, they did not fractionate out adherent cells ; , their results and ours correlated to a significance of p 106. Second, the statistical method used was a nonparametric rank sum test, better suited to the non-normal distribution of the data. Samples were analyzed based on their relative rank to each other, not on their absolute values. By this method, the relative ranks of the samples would not be expected to change very much even if the absolute values were to change. It is a member of the AAA family of hexameric ring ATPases, and it is highly homologous [14, 15] to the six ATPases in the 19S regulatory particle complex. We named it PAN, for proteasome-activating nucleotidase [14, 15]. Neither this proteasome on its own, nor PAN, nor the two together can efficiently degrade proteins. However, when ATP is added to the PAN20S proteasome mixture, they rapidly degrade unfolded proteins [15], such as casein, and also globular proteins, such as the green fluorescent protein, GFP provided it is linked to some short unfolded sequence; [16] ; . PAN thus facilitates the entry of these substrate proteins into the 20S particle, and together with the proteasome forms an ATP-dependent protease, with certain features similar to those that occur in bacteria e.g. they have a distinct ATPase domain or subunits, and a proteolytic compartment ; . Like them, its rate of ATP hydrolysis increases when it binds a protein substrate. Nadia Benaroudj showed that the PAN20S particle complex consumes approx. 300400 ATP molecules each time it degrades a protein the size of casein or GFP, which is about one-third as much as the ribosome uses in synthesizing such a protein [17]. Four functions have been proposed for the proteasomal ATPases: they facilitate substrate binding; mediate opening of the gated channel in the 20S proteasome to allow protein entry; are involved in substrate unfolding; and promote protein translocation [14]. We now know that PAN and the homologous 19S ATPases ; serve all these functions, and we are beginning to elucidate its ATP-dependent mechanisms in detail. Dr Benaroudj and myself were able to follow this unfolding process using GFP as a substrate, since it is green in the folded state [16]. By itself, PAN with ATP is able to catalyse the loss of GFP fluorescence. This `unfoldase' activity requires ATP hydrolysis, and represents a new kind of biochemical process. Another postdoctoral fellow in my group, Ami Navon, was able to show that unfolding occurs on the surface of the ATPase ring [18], but the mechanisms by which ATP-induced changes in its structure cause substrate denaturation are completely unclear. When the 20S proteasome was also present, PAN translocated the unfolded molecule into the 20S proteasome, where it was digested. Thus unfolding and translocation are normally linked processes. Recently, in collaboration with Yifan Cheng and Tom Walz, David Smith a postdoctoral researcher in my laboratory ; succeeded in capturing images by cryoelectron microscopy of the PAN20S proteasome complex, which associates in a transitory manner [19]. PAN appears as a `top-hat, tworing' structure at either end of the 20S proteasome. Thus its outer ring appears to bind substrates and to translocate them through its central channel in the 20S proteasome's outer ring. Furthermore, we found that images of PAN could be exactly superimposed upon the densities i.e. the two pseudorings ; in the base of the 19S component of the eukaryotic 26S proteasome e.g. as shown in images from Wolfgang Baumeister's laboratory ; . FLUOROQUINOLONE-INDUCED TENDINOPATHY: SUBJECTS AT RISK, PATHOPHYSIOLOGICAL MECHANISMS INCRIMINATED, THERAPEUTIC MANAGEMENT. Unfortunately, although these molecules are not associated with severe life-threatening complications, they have nevertheless been associated with tendon injuries responsible for functional disability. The frequency of these complications is probably underestimated. There is a variable lag-time 3 to 5 days ; between introduction of the antibacterial and onset of pain. The symptom most frequently reported is pain over the tendon affected and the tendons most frequently affected are those submitted to high constraints. Bilateral injuries are present in 66% of cases. These three forms of VEGF are abundant in human tissues 19 ; and are present in the human endometrium 20 ; . This is the first demonstration of VEGF mRNA in leiomyomata. Products corresponding to other VEGF species, VEGF , 20 ; and VEGF 18 ; , were not seen in this study. The presence of VEGF mRNA in myometrium and leiomyomata was confirmed by in situ hybridization. In myometrium and leiomyomata, VEGF mRNA seemed uniformly distributed throughout the smooth muscle cells. Proliferative phase myometrium showed similar expression of VEGF mRNA to secretory phase tissue. This contrasts with the changes in VEGF mRNA expression in human endometrium 20 ; . The punctate distribution of VEGF mRNA in proliferative endometrial stroma disappears in the secretory phase, whereas expression in glandular tissue increases during the secretory phase and is most intense in the glands of menstrual endometrium. Myometrium consists largely of smooth muscle cells and does not contain glandular tissue; thus, the in situ hybridization results showing no cyclical differences in VEGF mRNA distribution in myometrium or leiomyomata may be expected. VEGF-like immunoreactivity was also demonstrated in myometrium and leiomyomata, suggesting the presence of VEGF peptide in these tissues. The RNAse protection assay performed in this study is the first quantification of VEGF mRNA in the human myometrium. Increased levels of VEGF mRNA were found in the secretory myome?rium compared with the proliferative myometrium. This difference was small, only approximately 3-fold, thus explaining why a gross difference was not seen between proliferative and secretory phase myometrium using in situ hybridization. During the secretory phase of the menstrual cycle, spiral arterioles, which are the end-artery terminal branches of the uterine artery, develop from straight arteries in the myometrium and basal endometrium into the thickened superficial zone of the secretory endometrium 28 ; . At this time the endometrium is highly vascular, and the process of angiogenesis, with proliferation and migration of capillary endothelial cells, is undoubtably involved in growth of the spiral arterioles. VEGF is a secreted endothelial cell mitogen and angiogenic factor 19 ; , and thus increased levels of VEGF mRNA in secretory phase endometrium and myometrium may be expected to play a role in this angiogenesis. Cyclical changes in human uterine tissue are under the influence of ovarian steroids, and it is possible that the increased VEGF mRNA levels observed in secretory myometrium are mediated by estrogen and progesterone.
